You should keep the mind in binding affinities with measuring solvents. when stable, only then measurement should perform.

thanks for your answer. everything is stable, but i always find such result and i just need to find an explanation for that. why a dramatic decrease when there is not a great difference between 40 and 50 µg/ml of tested sample.
