I am going to analyse bacterial culture media for VOCs profiling. SPME is a used method for headspace VOCs extraction. Can I do VOCs profiling by using only one sample and one run GC-MSMS? Or I should run GCMS for VOCs detection and after that run GCMSMS for interesyed compounds?
While you are preforming MSMS you will not be able to monitor the peaks witch are eluting in the same time, In your example the chromatogram will be very crowdy. The combination of retention time and ms spectra will be sufficient to do the profiling, use the msms for the compounds witch you can not identify with the library.
GC-MS will provide the best global view (ie., profile) of the VOCs produced. If you decide on a specific metabolite you can then perform GC-MS/MS to increase specificity (SRM) and/or lower detection limits (SIM). You should be able to id the compound(s) via NIST mass spectral db search from the GC-MS analyses AND confirmation with true standards.
