We have some antibodies which can stain the cell membrane. What kind of experiments can I do to make sure the epitopes they recognize are from proteins? Thanks a lot.
Perhaps you can do a Co-IP experiment.
Compare your western blot with the stained protein gel in the same cell membrane prep.
Treat your sample with a protease, such as trypsin, to see if the antibody stops binding.
We have a human IgM antibody. It can induce cell death by CDC with rabbit or human complement serum. I am wondering whether mouse complement serum will also work?

I have a IgG secreting hybridoma. Unexpectly, when the cell number increase, the culture turn red and the cells are not happy. Any suggestions?

Im doing PBMC isolation -> CD14+ enrichment using magnetic beads -> stimulation setup. My negative control is just cells in cRPMI but they seem to get activated over and over again.

When you use RIA, with a control the unknown sample with antibodies, you bare in mind the binding sites of the antibodies. However you still need to measure labelled antigen (radioactive) and not...
