Spectrophotometrically derived pigment concentrations for available EPA standards agree reasonably well (+/- 20 % ) and results are corrected for the presence of pheopigments. But significant amounts of chlorophyll derivatives are present, pigment concentrations determined spectrophotometrically will be overestimated. Hence Chromatographic method is useful for easy solving the purpose.
You can try, first, with TLC or HP-TLC this a fast, cheap and versatile method, and you can made a quantitative analysis too. In the same way you can try with HPLC, exist many papers about this.
Use HPLC, start with high organic say 70% methanol or acetonitrile at 1 mL / min end gradient at 95%. Set absorbance detector at 440 nm. You should have no problems separating out the chlorophyls. Here is link to one of many hplc methods.
