I want to generate TR-beta overexpressed HepG2 stable cell line. For generating a lentiviral vector that have my interest gene insertion (TR-beta), that have puromycin or blasticidin resistance gene as selectable marker, and expressing GFP , what should I do? I dont know which Envelop plasmid, packaging plasmid and transfer plasmid are the best choice for my purpose.
I would suggest a VSVG-pseudotyped envelope plasmid like pMD2.G because you can use it for many other cell types in the future. As for the packaging plasmid, I would recommend a 2nd generation one like psPAX2 or pCMVR8.74. That way you have flexibility when choosing your transfer plasmid; can either be a 2nd or 3rd generation in this case and you don't have to add a fourth plasmid if it's the later. I think the smaller the transfer plasmid is, the better. Hope this helps and good luck!
