How can I calculate the dissociation constant (Kd) of binding from a cationic polymer-DNA complex in a gel retardation assay?

Yiou need to plot a saturation curve ( hyperbola curve) , i mean the plot of the concentration of the ligand that are bound to the receptor ( Y axis) as a function ot the free concentration of the ligand (X axis).. Fom this , you estimate the saturation , the assymptote Cmax on y axis. . Kd is the concentration measured on x axis that correspond to Cmax /2 (measured on Y axis ).
