The following document explains the method to calculate enzyme activity:

Also  Enzyme activity could be calculated using the following equation:

activity (U/L) = ((absorbance variation/time)/molar extintion coefficent * path length) * 1 micromol * (total reaction volume/total enzyme volume)

CALCULATIONS:

Standard Curve:

∆ A410nm Std = A410nm Std - A410nm Std Blank

Prepare a standard curve by plotting the ∆A410nm Standard vs the milligrams of Glucose.

Sample Determination:

∆ A410nm Sample = A410nm Test - A410nm Blank

Determine the milligrams of glucose using the standard curve.

Units/ml enzyme = (milligrams of glucose liberated)(5)(df)/((60) (1) (0.03))
