I have to find out the drug concentration in my formulation in which I added 15mg drug, for its, firstly I prepared standard curve of known concentration of drug and got beer-lambert law linearity after that to find out the drug concentration in formulation i dissolved prepared formulation in same solvent Ph 7.4 Phosphate buffer in150ml volume to get 100microgram/ml from this solution I made same concentration standard curve and now what should I do to find out actual drug concentration in 150ml solution when I will put the value of absorbency of sample formulation in known concentration of standard curve, suppose I get the concentration value “z” then I will multiply “z” to its Dilution factor then multiply by 150ml (“z” x Dilution factor x 150ml) then I will get the total drug concentration in 150ml Phosphate buffer solution in which I added 15mg drug which i have to find-out. Please tell me anyone.?

Plot calibration curve [concentration in mcg vs absorbance] use following eq.
Drug released in mcg = [Abs + intercept] / slope
Amount of drug in mg =
[Conc. in mcg x dilution factor x volume of dissolution medium] / 1000
Drug release = [Amount released x 100] / labelled claim
The term 1000 is used to convert microgram to mg.

You can get the data by using the formula
1. Conc of drug in Microgram=[Absorbance-Intercept]/Sloe from the calibration curve.
2. Conc of drug in Milligram=[Conc. in Microgram x dilution factor x volume of dissolution medium] / 1000.
3. % drug release= [Amount released in step 2 x 100] / labelled claim

What is your final formulation? as you are saying that you added 15mg drug in your formulation so before going to calculate drug loading (Total amount of drug in final formulation) you should separate the unentrapped drug
