does anyone have experience with TCR transcription upregulation? I would like to increase TCR mRNA amount in the cell in general, without specifically stimulating certain TCRs, but instead I would like to achieve overall TCR transcription upregulation (ideally in all cells in a sample). The stimulation does not need to be TCR specific, but it could also increase transcription levels in general. I would really appreciate the help because I could not find much in the literature.

In the aspect of lymphocyte differentiation, unless the cells are not exposed to certain antigens, they would not upregulate differentiation-associated gene transcription except for proliferation-related maintenance gene expression. However, PMA and ionomycin would permiate and activate second messengers or effectors in non-specific ways. You need to negotiate combined use of each concentartion without cell damage. If the cells are put in serum depleted (1~2%) medium for overnight, before to transfer to normal serum level for up to 6h, followed by the determined concentration of the two drugs. it would be an inductive cell differentiation.
