A question to those of you who use bacteria in order to produce reasonable amounts of bio macromolecules (e.g. circular double-stranded DNA molecules without any strand breaks, or proteins/protein complexes/enzymes, or inclusion bodies) in the quasi-natural state that are sensitive to shear forces, heat, chemicals etc.: How to you achieve lysis of the bacterial cells? I am looking for data illustrating the efficiency of the various lysis techniques. I would be especially interested in data related to the preservation of circular double-stranded DNA after lysis. I did meet people working in industry in projects in which intact DNA of this kind was needed, and they did not even have estimates regarding the ratio of well-preserved DNA molecules after cell lysis, relative to the number of these molecules in the intact bacterial cell. They were satisfied as soon as they had collected a - in their eyes reasonable - amount of these molecules. Would you assume that such a situation should and could be improved?
Alkaline lysis or any Tween treatment is much enough for bacterial cells lysis, or any slow enzymatic can do easy and effective lysis to get workable and intact biomolecules.
