I am going to perform a CRISPR/Cas-mediated knock-in. Unfortunately, a donor vector which I have has a C>T SNP right at the middle of one of the homology arms. The SNP is at position 400 whereas my homology arm is 840 bp long. It leaves me with remaining 440 bp in the HA. Should I correct the SNP before doing the knock-in?
