I would like to see by confocal microscopy the activity of the mitochondria potential membrane using TMRM. The problem is that I can't find a way to stain at TMRM in situ. I can process the tissue with a microtome and I can't fixed the cells
No It doesn't. If I fixe the cells, it will kill them somehow, and I won't be able to see the real potential membrane in mitochondria. I need to find a technique which doesn't need to fixe the cells.
I have already tried to cut the cartilage with a scalpel in small pieces and then stain them with TMRM but the problem is that the ends are always too large.
The technique of microtome is a good one but requires to fixe the cells. I would like to find something to by pass that and no fixe the cells, to preserve the tissue in his living status and then stain it with TMRM
