I have repeated the experiment twice (in triplicates/experiment) and have observed increase in viability with increased nanoparticle concentration (100 ug/ml-1 mg/ml). Can anyone please help me with the explanation or has anyone read/observed a similar trend somewhere?

How are you determining viability? It has been shown that some colorometric assays can give false positives because of interactions with the nanoparticles.

It is important to check several assays, and even noncolorometric ones.

Hi Ruby, all adherent cells, like fibroblasts are happiest when they can adhere tightly to a surface. If the NPs are providing a larger surface area for the cells to grow than a regular tissue culture plate, they will be happier still.
