I'm synthesising silver nanoparticles from silver nitrate with pomelo peels extract. Currently my UV-Vis results shows peaks ard 400-450nm. Other than using the UV-Vis to prove the presence of silver nanoparticles in my reaction mixture, I am in search of another method to prove the peaks from the UV-Vis is that of AgNPs.
You may go for ICP -AES or XRD or SEM for elemental analysis.AFM is also a good option.
Digest in nitric acid and add chloride to the solution. Most chlorides are insoluble but silver chloride isn't. Any precipitation or cloudiness at this stage is likely to be silver. Otherwise as others above suggest - EDX. I'd also add ESCA-XPS or Auger to the list.
The first question I should have added is the obvious one related to one used in bird identification. Is there any reason why the material would not be silver? You start with silver nitrate. What other possible metals could be produced?
You Can use XRD technique for confirmation of Ag nanoparticle formation. XRD is a fingerprint technique like IR. If your XRD pattern has broad peaks that will tell you that your material is Nano and the peak positions have to match with the peak positions of XRD pattern of Ag metal. TEM and Electron diffraction pattern is also useful for this kind of analysis.
From XRD analysis you can identify the type of silver nanoparticle produced by your green synthesis method. Through EDS analysis you can quantify the % of silver present in your synthesised silver nanoparticles.
Yes as others suggested, peak position in XRD pattern can identify the Ag nanoparticles. By EDX you can find the elements present in your sample. but I am curious to know Ms. Jyoti Gavali that how can we do elemental analysis by AFM.
If you do not have access to that analytical devices as described above you can try the very easy chemical way:
1. Centrifuge the nanoparticles and discard the supernatant.
2. Dissolve the particles in conc. HNO3
3. Make typical silver salt reactions like adding NaCl --> white precipitate; NaI --> yellow and so on ;)

Do a EDX mapping in TEM , as well as HRTEM to match the lattice plane of Ag, also XRD, Atomic absorption spectroscopy can be helpful
And Deepanjalee - what if they have none of these expensive acronym technologies in their laboratory?
If someone working on nanoparticles without using TEM , then i am afraid no one can help it .
I agree with "If someone working on nanoparticles without using TEM" - visualization is essential. However, consider the sample preparation and microtoming. I use one larger-scale irrelevant analogy here. Consider that you wanted to work out the proportion of the liver in the body. A vertical slice through the body would produce a totally different answer than a horizontal slice. Remember too that one is looking at a 2D representation of a 3D particle and shape is a 3D property.... Many parameters used such as aspect ratio are ratios of 2 2D measurements. So you still need to take care with SEM and TEM.
nanoparticles are zero dimensional unless it is a nanorod or nanowire (1D), nanofilms (2D)
Hi Deepanjalee I think you need to read your last response and alter it. A zero-dimensional object is a point with no space - a mathematical concept only. One could not examine a lattice plane in a crystal if your comment was right! Nanoparticles (as are all particles) are 3D....
nano particles have zero D bulk but have 3D in nano and nano particles are called three dimension quantum confinement.

The question was a simple confusion. The knowledge of basic stoichiometry is necessary for understanding such simple questions. No rocket science is required.
Why don't you just use XRD? if the particles are silver it should be a simple XRD pattern to find. if there are other elements in nano particles then the pattern should change some. So long as they are in the structure and not on the surface of your particles .

Why did not you go for EDX, XRD or many other techniques? Tina is also rigth and Mr Alan has also explained clearly. Is there some technical problem or some other experimental issues? Can you explain please?

Alan F Rawle. Hi sir. Good day to you. I apologise for the late reply. I used to collect stamps too when i was young but I'm not sure if I have any collection from Colombia. Perlis is the smallest state in Malaysia and its pretty low profile so that might explain the lack of Perlis stamps distribution and its price. =D hope you've been well since your last comment.
XRD and ICP are two of the techniques to confirm the formation of AgNP.
