Is is possible to find the LDH concentration using a solution NAD+, LACATE and CAPS buffer with the LDH sample at 340nm to find the LDH concentration in human blood? what would be the protocol?
I think that you need commercial kits to meausre the concentration of LDH. The materials you have mentioned are required to measure the activity of LDH not its concentration in a sample. However, you may be able to estimate the concentration of an unknown sample if you could have LDH with known concentration and compare their activitities. On the other hand, as 340 nm is used to treace the conversion of NAD to NADH, you should make sure no ther enzyme in the assau coktail has such function.
