Hello everyone, I just changed labs, and althought the electrophoresis source is the same as the old ab (EPS 601 Amersham), I can´t seem to be able to change the limiting parameter to current, it´s set on voltage and I have no idea how to change that. I always worked in mA, so I have no idea what voltage for a mini gel I should apply.
I´ve put 200, but then I only get 1.5 mA for the gel, which seems to little and will take forever to run... I used to use 45 mA to run in about 1.5 to 2 hours. Anyone have a tip for this? What voltage is adequate, and is the low mA ok for this?

What kind of gel are you running (e. g., 1D- or 2D-PAGE or agarose gel)?

Usually I use 35mA per minigel and 200V. Intensity is limiting at the beginning of the run (voltage being around 180V) and then voltage becomes limiting later due to the heating. Run is completed within one hour.
Hello everyone, I edited the question, it´s a 601 source, not 3501. I´m running a min SDS-PAGE 0.8mm thick gel.
The thing is, I would like to set 35 mA per gel and stick to that fixed value, while power and voltage would vary. I used to do this at my old lab, but the underlined parameter here in my new lab is the voltage, not the current. So, If I put 35 mA per gel, the voltage goes to the setted value, and the current does not reach my input value... does that make any sense at all??

The underlined parameter is actually the one limiting for the run so what you have to do is to set higher values for both voltage and power so that current becomes finally the limiting factor (and get underlined when running is on).
hello Dominique, I did set both voltage and power to their maximum settings, but the current did not show any underlined... I did have a small issue with the buffer... perhaps redoing the run in these conditions the underline will show up?
Hi again, normally current should be limiting... Check the plugs, électrodes and buffer. Normally, 35mA should be reached for a voltage around 200V in Tris Glycine buffer.
