I am doing phytochemical analysis of some plant leaf extracts. But I have a doubt. I am doing quantitative analysis of saponin where I take "Quilaja Saponin" as standard. But as per many Saponin protocol the colour should come yellow, red etc. I am confused as I don't get this colour during my experiment and also there is no proper explaination regarding colouration of Saponin. I am using Vanilin Sulphuric acid method.So can anybody please clear my doubt?
Madam, I prepared plant leaf extracts using water. However, for Vanillin- Sulphuric Acid Method, 8% vanillin is prepared in 99.9% ethanol.

color
2. 1% solution of vanillin in a mixture of sulfuric acid and 96%
ethanol in a ratio of 1: 1 - The spots are slightly purple
3. 1% solution of vanillin in a mixture of ethanol and 3% chloride
acids in the ratio 1:1 (Reagent Godin [4]) - Spots transparent colorless
4. Antimony(III) chloride solution - No saponin zones were found on the chromatogram
5. 01% resorcinol solution in 10% ferric chloride solution
(III) (Reagent Bial [4]) --The spots are slightly colored brown
6. 20% solution of phosphotungstic acid in
95% ethanol - Spots are light purple
7. 20% solution of phosphomolybdic acid in
95% ethanol - Dark green staining of spots on a light greenish background of the sorbent
