Hello, how could I transfect cardiomyocytes with siRNA and get the target efficiently?

1- After 9 days of siRNA transfection, I'm required to gather samples, however I'm not sure if this is okay or if the effect of the siRNA would be weakened.

2- Which is it better for CMs, reverse or forward transfection?
