I plan some experiments over the adipose tissue immune cells in mouse fat, but can't find out: How many animals should I use to generate reasonable cell numbers for analyzes.
In normal adipose tissue, there should be practically no inflammatory cells. However, if there is necrosis due to trauma, there will be macrophages and a few lymphocytes. I agree it depends on the mouse type and that normal mice fat will not provide you with top macrophages and T cells. To optimize on the matter I suggest ob/ob mice. They are severly obese, insulin resistant and glucose intolerant. Their inflamattory pattern (including in adipose tissues) should offer you enough various lymphocites, with «n» just a few mice. You would like to test on visceral fat first. Our paper here will give you some pointers, even if we do not specificcally investigate lymphocites. But you can see there was apoptosis and fibrosis in the cardiomyocytes, conditions that can not occure without a subchronic inflammatory background. The macrophages in ob/ob mouse adipose tissue is due to trauma and fat necrosis, not putative inflammation due to hyperglycemia, obesity, or glucose intolerance. This is a misconception which must be corrected in the literature. Again, I urge you to read my paper and familiarize yourself with fat necrosis, including the histopathology.
