I am just wondering if anyone has an experience with touchdown PCR during their site-directed mutagenesis experiments.

Based on my past conventional PCR experiments, I do not see my target bands on the gel. I think that I have to change my PCR strategy possible due to poor primer design.

While I consider redesigning my primers, I also want to ask for your opinion on using touchdown PCR for mutagenesis experiments.
