Have You ever faced problems with atherosclerotic plaque staining? Yesterday I stained mice aortas full of plaques. Very few of them are not stained. So then I repeated the whole process again, but the results are the same. Do you have any advice?

My ptotocol is as following, after aortas fixation in 4% PFA (in PBS) O/N at +4°C.
I wash aorta 2x 5’ in PBS. Afterwards I rinse with 60% isopropanol.
Afterwards I incubate in ORO solution O/N at +4°C. Next morning I rinse aortas again with 60% isopropanol. Then I wash aorta 2x 5’ in PBS.
That's it. 22 aortas are fine, 4 aortas are not stained. Have you ever had yhis kinda problem? Have you figured out what is wrong? And is there any possibility you can stain those plaques again?
