This may not be the answer you want, but I have never had success using a turbo blotter with high molecular weight proteins (even using the specific programs recommended by the company for high molecular weight proteins). I personally recommend going back to the old school slow wet transfer. It may take longer to run, but it will save you a lot of headaches in the end instead of trying to optimize the turbo blotter. Plus, the quality and efficiency will be far superior.

For 450k Da mass means, possible to use fast  turbo transfer for transfer but the final result not fair., because protein in higher MW means have so many intraction so the instrument have difficulty to read and process it so we get problem.,

So dont use turbo transfer for this condition., But the instrumental manual they are saying up to 600k Da also able to transfer but in practical we got difficulty.

Slow wet protein transfer is very best but take time.

Now one HC Turbo transfer introduced exspecialy for protein transfer, i have not worked yet, but i heared it's very fast and user friendy, digital monitering option also there like this. Try.,

This is major practical problem.,  Bio Rad turbo transfer means above 150k Da, not possible to do efficiently, but somebody said to us it's possible, but i have experienced, not at all possible., 

I have used protein 530k Da with High CDefinition turbo transfer, its recently introduced, it will make faster., so you try this.,

Hi, thats  a good point. But some papers have shown it around 450kDa probably after digestion to remove glycosylation.
