We recently acquired a CHO line from Sigma (00102307) adapted for growth in suspension in Sigma CHO Protein Free Medium. The cells are pink and not growing. Has anyone experienced the same?
It would be also advisable to check for CO2 level in the incubator. Sometimes, the actual level of CO2 may be lower even though a reading of 5% displayed.
Check whether your media is having pH indicator. If so pink colour of the media indicates alkalinity. Check your CO2 incubator by keeping some other standard media.
Hi Ana,I agree with the previous suggestions and probably you have also to determine the right density of cells in the volume of medium you use, in order to promote their correct growth.
HI Ana, the only suggestion I would add to the previous is to check that you are adding the NEA (non essential aminoacids) to the culture media. I never used CHO in suspension, but the adherent one do not grow without NEA.
