We've had good luck with this method using human peripheral blood neutrophils, but not with mouse peripheral blood or thioglycollate-elicited neutrophils.
We have used another substrate, TMB, tetramethylbenzidine I believe. It has worked pretty well since I can remember. It could be just a question of substrate concentration too, maybe you're just at the borderline for the test.
Also, are you getting enough neutrophils from the other preparations? Try to raise the cell number to increase the signal and you might find something. Good luck!
You can try with Amplex Red as fluorogenic substrate, but in complex media, I don't know if it will be specific. Or see with immunological method as ELISA or SIEFED if you have antibodies against mouse MPO. Good luck!
