I am trying to get CRISPR/Cas9 working in my lab. I have a strain of Aspergillus that expresses the Cas9, and I need a way of introducing the gRNA.
I think CRISPER/Cas 9 works better with prokaryote rather than a eukaryote.
You can try endogenously expressing systems. One example is pCRCT plasmid that allows you to express the sgRNA and Cas9 in one plasmid. It works fine for yeast systems but not sure for fungi. Here is the link to the paper:
