We are trying to see how the nuclear translocation of NF-KB is affected in 293T cells by different treatments, and we recently purchased this kit from Millipore. However, we are having contamination problems of nuclear fraction into the cytoplasmic fraction (we are detecting histone 1 equally in the cytoplasm and nucleus). I was wondering if you had ever used the kit and/or if you have suggestions to prevent contaminations from other fractions.
