B16 murine melanoma cells tend to show higher potential of melanin synthesis in 3D culture than in 2D culture. Matrigel might influence on the plasticity of the malignant melanoma phenotype characterized by melanin synthesis (differentiation) and the invasion into ECM (the production and secretion of MT1-MMP and ADAM family).
I recommend you to perform the experiment using both B16(F0) and B16(F10) cells.
I would not use the B16 (whatever "F") model s.c. because it gives highly necrotic tumors with difficult data to interpret.
This is actually the case with B16F10 (Darro et al., 2005, here attached) and I do not share Go's advices.
B16F10 cells are much more interesting in terms of cancer biology if injected i.v. to give pseudometastases into the lung (see the attached articles by Mathieu et al., 2007, 2012) or in carotids to give pseudometastases to the brain (see the third attached article).
