Has anyone tried metabolically labelling HEK293 using 5- ethylyne 2-deoxycitidine or deoxyUridine. I am trying to label the cells but it's not working, it doesn't seem that there is effecient incorporation. Has anyone ever tried HEK to do DNA labelling and then click-reaction using Cu(I).
I am kinda confident that it's not the click-reaction step. I have tried about 15 different conditions with different buffer, with/or without ligand, 2 different flourophores-azide but still a very low labelling that you make you think it's just a background of the dye.
