I'm seeking advice on expressing and purifying IgE antibodies in a prokaryotic expression system, specifically E. coli. I'm interested in hearing about successful strategies for achieving high yields.
For my purposes, post-translational modifications are not a primary concern. I've come across studies on IgG expression in SHuffle cells, but the reported yields seem modest. I'd love to learn from anyone with experience expressing IgE in E. coli.
Hello Solomon, you can try growing E. coli in Dynamite media. You can achieve high optical densities and, as a result, higher protein yields. I’ve increased the yield for some of my proteins 10-fold using Dynamite media. See the publication below. Good luck!
