I am interested in testing several chemical compounds that might have an effect on endothelial adhesion and/or transendothelial migration of monocytic cells. Since the ultimate aim is to test the compounds in vivo, I would like to use mouse or rat cell lines.
I have heard, that it is rather difficult to perform such assays with cell lines. Does anyone have experience with this?

I think you will find this type of assay very difficult to perform. I would say impossible. Most monocytic cell lines are constitutively adhesive and will not behave as primary cells in transmigration assays. You might try WEHI3 cells, which are not very adhesive, but overall my advice would be to avoid this difficult assay using cell lines that probably won't work.
