We are trying to obtain mouse blood to isolate PBMCs and do some tests. We do need the maximum amount of blood we can get but cannot use the mice as long as they are alive. We usually do cardiac puncture under anesthesia but for now we do not have approval for that. We tried cardiac puncture on CO2 sacrificed animals but have problems with coagulation, no matter if we add heparin or use EDTA coated tubes. Does anyone has some suggestions how to prevent coagulation?

Although you still need them to be alive (anesthetized), the best approach to collect blood from mice is probably by eye removal. It is not pleasant to perform, but is quite easy to learn and allows for the collection of high amounts of blood. I am not sure if this approach will be useful for you in this particular situation, but it is worth trying to help you.
I know that is best when they are anesthetized. But now the requirement is that they are dead.
