Has light sheet mic imaging capacity the chromosome? I am planning the 3D -FISH experiment with light sheet mic but I am not sure that the instrument can visualize the targeted genes on chromosome. Have you any idea?

I am uncertain what you would determine by using FISH to identify genes on a chromosome in 3D ? In that context FISH is used just to identify the gene position in 2D i.e along the length of the chromosome, doing this in 3D would add little. If you are talking about looking at gene expression on a tissue specific basis (in whole mounts) the working in 3D would make sense. I guess I do not understand the experiment you are trying to do.
