The FLAG tag is short and shouldn't have any effect on H1R folding/activity. Be careful to insert sequence coding for FLAG downstream signal peptide sequence.

I'm not a specialist of receptor but it has to be targeted to ER membrane during translation in order to insert into it. For this purpose, the translated polypeptide might present a specific sequence at its N-terminus. But it is not compulsory.
