I have used Promega RNA kit, but it didn't work. The volume of CSF that was used was 6ml.

i don't know the Promega Kit but there are other good Kits around for CSF RNA extraction! I can recommend the miRNEASy Serum/Plasma Kit from Qiagen! Using 200-250 µl CSF is enough to isolate about 10-30 ng/µl RNA (purity ratios range from 1,4 - 1,6). In comparison to other Kits like miRVANA Paris or Exiqon Biofluids or the Zymo Kit, the Qiagen Kit gives the best RNA yield! What does "did not work" mean? No yield at all? What did you use for RNA quantification? NanoDrop? Bioanalyzer? Qubit? And what is your intention with the extracted RNA? You can also add Glycogen to your protocol as a co-precipitant to increase RNA yield! Hope this helps!

Thank you Johannes for you answer. Exactly, no detection was seen at all. Are you suggestiong to use qiagen kit or miRNAEASy kit? which of them? I used nanodrop for RNA quatification and I like to make cDNA from RNA and eventually run RT q-PCR.
