I work with fungi, and I failed twice to amplify ITS and LSU, following exactly the specifications of the manufacturer....

Did you try with these genes? do you have the programs for the thermocycler to chek if the temperatures I am using are fine?

I will try to contact technical support, but directly in Estonia.. here in argentina I donn´t knowif the seller knows about that

sometimes if you change the mastermix supplier it is necessary to change annealing temperature, it's depends a lot from the manufacturer and thermalcycler.

We have good results working with mixes for lighcycler but also for quite different cyclers as Cepheid SamartC

Hi Simon Robberts on, I tested this same Mast Mix this week and it didn't work too. With a kit from another company I got some results. I don't know wuat could be the problem.
