To date I have not been able to find any literature on the topic other than exogenous expression of SV40 T-antigen was ineffective. Has anyone attempted exogenous hTERT expression or a viral method? Any other suggestions would be appreciated. I am trying to isolate monocytes for functional assays that may require follow-up assays over the course of weeks/months.

Do you mean immortalisation of B lymphocytes? If yes, use Ficoll to isolate the monocytes and the B95-8-EBV + cyclosporin (1ug/ml, Sandoz) for transformation. For my knowledge this is the first description of this method: Neitzel H.Hum Genet. 1986 Aug;73(4):320-6.
