Since some time I've been getting quite a few droplets in my NTCs that have high and similar ch1 and ch2 amplitude, i.e. they find themselves on the diagonal of 2D plots. It didn't used to happen even with the exact same mix of primers and probes (I'm using probes, not EvaGreen).
I tested the water, the TE buffer (used to resuspend probes and primers), thested a brand new batch of primers and probes, different lots of cartridges, different lots of DG oil, pipettes and 96-well plates.
It also seems that the overall number of droplets has decreased over time. It also seems like the less total droplets an NTC well has, the more diagonal false positives it has.
Bio-Rad's manuals say that you can get diagonal false positives if you "mishandle" the droplets, but they never explained what "mishandling" means.
Any advice would be appreciated!
