I am starting to work with THP-1s differentiated into macrophages and wondered if I must differentiate with PMA every experiment or if I can culture and passage the already differentiated cells. Has anyone tried doing this? If so, which conditions worked?

After differentiation with PMA, THP-1 macrophages do not divide and multiply. because of this, you can not culture them. Every time we need to differentiate them and use them for experiment.

Hey, proliferation is restricted post differentiation although you can further differentiate the cells to M1 or M2 macrophages. Hence PMA differentiation has to be fresh every time.

You need to differentiated each time for experiments. Can't maintain THP-1 derived macrophage.
