I am using AML 12 cells (liver cell lines - non cancerous ) . I observed a change in their ploidy over late passages. Is it observed by anyone else also or it might be due to experimental error on my side ?
(I Have followed simple PI staining protocol as always ) .

Goodwin .. I used the DMEM/F-12 : A 1:1 mixture of Dulbecco's modified Eagle's medium and Ham's F12 medium with 0.005 mg/ml insulin, 0.005 mg/ml transferrin, 5 ng/ml selenium, and 40 ng/ml dexamethasone, 90%; fetal bovine serum, 10%
That seems ok as per ATCC

Do you use new flask every time when you subculture the cells?
It could be the characteristic of these cells I guess. Check the literature for people who published using this cells, and email the authors asking, whether they noted a similar issue. If not, go for a fresh stock from ATCC or STR fingerprinting to make sure your source is ok.
This is okay because AML12 are hepatocytes. Significant part of hepatocytes has 2 nuclei.
Does anyone know about the STR fingerprinting information about AML12 cell line from ATCC?
