I want to measure chlorophyll fluorescence in relation to the Cadmium stress in rice plants by using above mentioned leaf fluorometer. What is the correct way of doing this in order to take a better measurement? Is dark pulse compulsory to do this? Can't I use full flashed condition to measure this?
The system allows gas exchange and electron transport efficiency by a modulated fluorimeter analogue to PAM Walz. Usually you take a light curve by increasing light on dark adapted leaves, and collect fluorescence parameters under progressive toxification. In the field you may dark adapt with aluminium foil wrapped around a attached portion of flag leaf. after measurement take off the sample and measure Cd. I plan on using our LICOR 6400XT coupled with a PAM 2500 probe attachment on a 2x3 LICOR chamber built for the purpose soon. This technique is ideal for combined gas exchange and fluorescence measurements to get non-invasive insight into biochemical and physiological information on photosynthesis under stress such as due to light, salinity etc.
