I'm planning on doing a biocontrol study on Erysiphe alphotoides/ syn. Microsphaera alphitoides/ Powdery oak mildew, but first I need to isolate the fungus. Perhaps someone knows what sort of medium is best suited for this fungus?
The fungus Erysiphe alphitoides is an obligate fungus. So it can not be isolated on the artificial media in the laboratory. Therefore, you must complete your experiments on the living plant host (oak).
All powdery mildew diseases are caused by obligate fungi like Erysiphe, Uncinula, Leviellula, Phyllactenia, Spodoshaera, Microsphaera etc. All these pathogens are obligate parasites and they can not grow and reproduce far from the host plants.
Even you add oak leaves to the medium, this obligate fungus can not grow, because this fungus can not grow easily far from the living host plants. The obligate parasite fungi like the causal agents of powdery mildew can not grow on the different artificial media.
