Hi everyone!...

I'm trying detect NO2- on J774A.1 macrophages using Griess Reagent Kit for Nitrite Determination of Molecular Probes. However, I can't detect NO2-. I seeding 1e5 cells/well and 24 Hrs after, I add 50 ng/mL of LPS and them (24Hrs) I take 100uL of supernant and add 50ul of Reactive A and 50 ul of Reactive B of the kit... This is incubate for 30 minutes and after I read to 562nm... But I don't have differences between controls (W/out LPS) and treatments... Can anyone help me?
