Greiss reagent does not exist. The correct spelling is Griess. Griess assay is designed to measure nitrite but not NO. Under certain conditions NO is oxidized by O2 to form exclusively nitrite, which can be detected by Griess reagent. The protocol to measure nitrite is pretty simple and described in numerous papers
