As in the title...a membrane associated protein fused with GFP (target protein is expressed in nervous system). I guess signal peptide is not functioning as normal.. homozygous of GFP tagged protein is lethal for flies (stop development before hatch). But in embryo, GFP expression seems like in nervous system. Have to further to see whether they go to the membrane. Anyone has some comments on that?
If I understand correctly that your GFP is before the signal peptide, that will not work. The signal peptide must be the first thing that gets translated, in order to insert in the membrane at all. It will probably just aggregate in the cytoplasm or get degraded.
Thanks John! I have the same thoughts... Cause it is from one of my collaborator, they try to do the same tagging in flies as I did in C. elegans. But I put GFP after signal peptide... So, you mean... GFP before signal peptide probably disrupt the original signal peptide function?
It will most likely disrupt the function of the SP. In classical secreted or targeted proteins the SP is at the N terminus of the protein and gets most of the times cleaved after translocation/targeting.
