Currently I am treating HCC tumor cell line in vitro using my special compounds. I am doing kinetic cell cycle experiment and I found that my drug gradual induce G2M block after 8-16hr treatment. But after 24hr there is a general release from the block. Is there anybody have the same experience and what should be the potential reason for this?

Cellular cycles of tumor cells are one of the mechanisms for avoiding the action of cytostatics. Please see two articles that will help you.
How do you determine G2/M block ? There is a phenomenon, called "mitotic slippage", which might be the reason for your observation. But it really depends on how you analyze the cells. For example, if you are doing FACS with phospho-H3(S10)-antibody to see mitotic enrichment, then this staining will disapper after mitotic slippage, but the DNA content (propidium iodide staining) should remain unchanged to mitotic cells (4n), since the cells are not divided.
Also it could be that your compound is unstable in culture media leading to transient cell cycle arrest. Try to change media after 16h to see whether you can prolong arrest or not.
