I am looking for CHD7 silencing siRNA. I found one link in Santa Cruz Biotechnology as CHD7 siRNA (m). I need an opinion for what is the best company to procure this siRNA.
There are many vendors. I have used siren from dharmacon and qiagen. I generally procure the lowest available quantity. I didn't use pool, but buy them individually and test their knock down efficiency.
The company siTOOLs located in Munich sells customized RNAi reagents. SiTOOls Biotech has developed so called siPOOLs, which are complex mixtures of 30 carefully selected siRNAs all targeting the same gene. By this each individual siRNA in the mix is present at substantially lower concentration (1/30 of the entire pool - i.e.: if you tranfect at 3nM fc, each siRNA in the pool will be present at 100pM)
This results in 2 fundamental advantages: better knock down efficiency, because the 30 siRNA work cooperatively and published data indicate strongly that with as little as subnanomolar concetration of teh entire pool you will get KD >>80% (20pM per each siRNA in the pool)
More importantly you overcome off target effects. So the phenotype you get in fact relates to the gene you have been knocking down. Which is NOT the case for normal single siRNAs or the so called smart pools. Thats why you have to test so many different siRNas - and ideally do the rescue experiment. There is ample literature on off target effects - seed sequence binding and there is also a publication that highlightes the siPOOL idea.
