For long term storage it is better to store sample in RNAlater at -20 °C, but yes, you can store it at -80. Just be careful for the formation of crystals int he frozen sample, to avoid it you can put your sample in RNAlater overnight, remove it from RNAlater and store it at -80.
About RNA quality i always extracted good RNA from RNAlater treated samples, both stored at -20 and -80. But personally i prefer to extract RNA instead using RNAlater (if possible), it is always a better solution. I use RNAlater just in "extreme" cases, such as samples collection on the field or samples transport.
so if samples were stores already in -80 with the RNA later on, does that affect the RNA later ? Should I remove the RNA later now and freeze them again in -80 ?
