I really need help. I loaded my uncut, restriction digests, and PCR on my gel, and when it was done and I put it under the UV light, I realized that I forgot to load a ladder to measure my samples. is there anyway I can measure the samples without using a ladder? I'll add a picture, my samples are on the bottom half of the page.
Is there anything on the gel that you know the size of? Normally most experiments should involve controls where you know something. Use that as a proxy for the ladder. (this might not be a publication grade figure, but at least it will help you figure out what's going on for yourself)
your samples are very concentrated so you could load much less on another gel with a size standard. If you have loaded most of your sample then pick identical samples from this gel and pool them . then run the pooled samples against a size standard ( ladder) then when youknow the size of most of your samples use them as a ladder and read off the unknowns either by eye or by plotting 1/s where s is the distance run against the known sizes and read off the unknowns against this nearly linear graph
That gel is a toast. The best way is to try another gel with a MARKER.

From current gel, you won't have a precise way to figure out the sizes of the DNA fragments. If you don't have DNA or colonies any more, I would cut out each lane's bands and gel-purify them. Then, run them again on a gel with marker. But, it is a bit late to do that now. If you still have DNA or colonies (w/o DNA), you should start it again.
if you have any of these product left, you can run it with a marker and than you can predict the others. If you dont have any unloaded product. You can gel purify one of them and you can load it with a marker to predict others.
