I added 1%dmso and 99%PBS to my compound which made suspension rather than solution. Can I still use this for MTT assay? Even some of my compounds precipitate on PBS addition, what to do with that?
I gave an explanation to some question in this field (topics) but I will tell you again. What I do with highly lipophilic compounds is dissolve them in 100% DMSO (certain conc. ca 10-4 M) and then the concentrated solution I dilute with cell culture medium (10 uL in 990 uL) thus making the DMSO conc in medium low and substance dissolved (not suspension). However I cant be sure that it will work with every substance but you can try it. Also after that step i mix 100 uL of cell suspension with 100 uL of dissolved substance.
You can try by increasing the concentration of DMSO, (better to use 100% DMSO as suggested by Nikola M Stojanovic). However, if the compound is getting precipitated upon addition of PBS, i think it wont be suitable for cell culture.
