Can anyone help me with the information about how long is the NDV virus isolated from the allantoic fluid of embryonated Chicken Eggs will be stored at -80C?
No is isolated from embryonated chicken eggs (9-11 days old). After when we harvested the allantoic fluid we then put the virus in -80C while waiting for the rest of our materials to arrived for further clarification and purification process but we are not sure for how long the virus (NDV) can stay at that -80C. Hoping it won't have effect on the virus for long storage like say for 3 Months?.
I am not an expert on NDV. But what my friend who is a avian disease specialist told me is, Allantoic fluid containing Newcastle disease virus can be stored for up to 6 weeks at 4°C without significant loss of titre. For longer duration of storage stabilizers gelatin and skimmed milk powder are being used.
Store NDV at -80 I dont´t know, but at -20oC in allantoic fluid is OK for at least 2 years! But, if you need have perfect control of HA title, be carefull and preserve the virus in some aliquots to use along the time...You need to do another smallest alliquots only to measure HA title along the time of project... virus stored with proper media (like allantoic fluid or cell culture media) no problem...but must use the proper method....even u can store one or two allique in the liquid nitrogen for long term use....but do the TCID50 before storage.
You can store aliquoted NDV in allantois fluid, at -80 C, very long time (over 10 years) without any problem, about HA titers, especially velogenic viruses. You may have any problems with infectivity, if virus stored more than 7-8 years.

i,ve got velogenic 1972 ND seed in alantoic fluid stored at -80C & they,re still work well. as long as they,re in whole virus form. It will be better to aliquoted them in several microtubes before you stored them. Those microtubes could be use to check the HA titers & infectivity every year or 2 years.
As long as your freezer doesn't fail, the NDV should remain viable. I have had several strains of NDV stored in 1 or 2 ml aliquots in sealed tubes for many years. Periodically as needed, I have thawed NDV and it replicates just fine. Just avoid repeated freeze/ thaws, keep the temp around -80C or lower, keep samples in nearly full tubes to avoid sample dessication and it will last a very long time.
As long as there is no freeze-thaw cycles, your viruses will preserve for years and still be infectious. This is not an issue.
